چکیده

Gas-processing metalloenzymes are of interest to future bio- and bioinspired technologies. Of particular importance hydrogenases nitrogenases, which both produce molecular hydrogen (H2) from proton (H+) reduction. Herein, we report on the use rotating ring–disk electrochemistry (RRDE) mass spectrometry (MS) follow production H2 isotopes produced deuteron (D+) reduction (HD D2) using [FeFe]-hydrogenase Clostridium pasteurianum, a model hydrogen-evolving metalloenzyme. This facilitates enzymology studies independent non-innocent chemical reductants. We anticipate that these approaches will be value in resolving catalytic mechanisms H2-producing design catalysts for N2 fixation.

Protein analysis by hydrogen exchange mass spectrometry.

Mass spectrometry has provided a powerful method for monitoring hydrogen exchange of protein backbone amides with deuterium from solvent. In comparison to popular NMR approaches, mass spectrometry has the advantages of higher sensitivity, wider coverage of sequence, and the ability to analyze larger proteins. Proteolytic fragmentation of proteins following the exchange reaction provides moderat...

Electrochemistry-mass spectrometry unveils the formation of reactive triclocarban metabolites.

Triclocarban (3,4,4'-trichlorocarbanilide, TCC) is a widely used antibacterial agent in personal care products and is frequently detected as an environmental pollutant in waste waters and surface waters. In this study, we report novel reactive metabolites potentially formed during biotransformation of TCC. The oxidative metabolism of TCC has been predicted using an electrochemical cell coupled ...

Perchloroethylene Analysis by Chemical Oxidation and Determination of Intermediate Products by Gas Chromatography, Mass Spectrometry

Abstract Background and Objective: Perchloroethylene (PCE) is a chlorinated hydrocarbon used as a solvent in many industrial processes. In contaminated water and soil a great deal of PCE is found. This study aimed to determine the rate of decomposition of PCE occurred after advanced oxidation. Material and Methods: In this descriptive-analytic study conducted (2011) in public health faculty...

In-Depth Characterization of Protein Disulfide Bonds by Online Liquid Chromatography-Electrochemistry-Mass Spectrometry

Disulfide bonds are an important class of protein post-translational modifications, yet this structurally crucial modification type is commonly overlooked in mass spectrometry (MS)-based proteomics approaches. Recently, the benefits of online electrochemistry-assisted reduction of protein S-S bonds prior to MS analysis were exemplified by successful characterization of disulfide bonds in peptid...

Characterization of hydrogen bonding motifs in proteins: hydrogen elimination monitoring by ultraviolet photodissociation mass spectrometry.

Determination of structure and folding of certain classes of proteins remains intractable by conventional structural characterization strategies and has spurred the development of alternative methodologies. Mass spectrometry-based approaches have a unique capacity to differentiate protein heterogeneity due to the ability to discriminate populations, whether minor or major, featuring modificatio...
